Differential responses to selenomethionine supplementation by sex and genotype in healthy adults

A year-long intervention trial was conducted to characterise the responses of multiple biomarkers of Se status in healthy American adults to supplemental selenomethionine (SeMet) and to identify factors affecting those responses. A total of 261 men and women were randomised to four doses of Se (0, 50, 100 or 200 μg/d as l-SeMet) for 12 months. Responses of several biomarkers of Se status (plasma Se, serum selenoprotein P (SEPP1), plasma glutathione peroxidase activity (GPX3), buccal cell Se, urinary Se) were determined relative to genotype of four selenoproteins (GPX1, GPX3, SEPP1, selenoprotein 15), dietary Se intake and parameters of single-carbon metabolism. Results showed that supplemental SeMet did not affect GPX3 activity or SEPP1 concentration, but produced significant, dose-dependent increases in the Se contents of plasma, urine and buccal cells, each of which plateaued by 9–12 months and was linearly related to effective Se dose (μg/d per kg0·75). The increase in urinary Se excretion was greater for women than men, and for individuals of the GPX1 679 T/T genotype than for those of the GPX1 679 C/C genotype. It is concluded that the most responsive Se-biomarkers in this non-deficient cohort were those related to body Se pools: plasma, buccal cell and urinary Se concentrations. Changes in plasma Se resulted from increases in its non-specific component and were affected by both sex and GPX1 genotype. In a cohort of relatively high Se status, the Se intake (as SeMet) required to support plasma Se concentration at a target level (Sepl-target) is: .

Postmeiotic segregation (PMS) of genetic variants occurs when a DNA heteroduplex formed during meiotic recombination goes undetected by repair enzymes and is transmitted unresolved to the meiotic products. PMS provides an alternative explanation for the origin of mosaics now attributed to half-chromatid mutation. In multicellular diploid eukaryotes, PMS could result in mosaic individuals with unusual migration patterns for proteins studied by gel electrophoresis. If the gonade were mosaic, complex progenies containing as many as six phenotypic classes at a single locus could be produced.

The pre-treatment of aluminium surfaces prior to adhesive bonding and coating processes is of major technological significance. However, the two major commercial pre-treatments, chromic acid and phosphoric acid anodising, both make use of Cr(VI) containing solutions with the attendant problems of waste disposal. The disposal of this harmful solution poses severe environmental problems. Therefore, other environmentally friendly alternatives are being investigated, one of the most popular candidates is the use of one-step pretreatment processes based on organosilane adhesion promoters.
This study examines a model system made from polished aluminium sheet treated with an organosilane; γ-glcidoxypropyltrimethoxysilane (GPS). The interface was characterised by TEM carried out on microtomed sections. The information gained from imaging and Electron Energy Loss Spectroscopy (EELS) analysis revealed a layer of oxide, approximately 70 nm thick. Silicon was found to have been incorporated within this oxide layer. Thus it is suggested that during the curing of the GPS, at elevated temperature, growth of aluminium oxide takes place which gives rise to an Al/O/Si phase.

Teachers' comments in the childhood school records (grades K-12) of 143 psychiatric patients and their matched controls were coded along 23 bipolar dimensions. Two methods of grouping these scales were compared: rational clusters and factor analysis. Factor analysis yielded more numerous and narrowly defined behavioural groupings. Schizophrenics, personality disorder patients, neurotics, and depressives were compared to their matched controls on each of the cluster and factor scores. Both schizophrenics and personality disordered patients were significantly less agreeable in childhood than their respective controls. Pre-schizophrenics also were significantly more unstable. Depressives were more independent than their controls, while neurotics did not differ significantly in any respect from normals in childhood. The data suggest that schizophrenia may have specific developmental patterns of possible aetiological or early diagnostic significance.
